[Greenbuilding] best energy-efficient setup? - thermostat on unused floor

How many returns and where are they located? You only stated where supply vents are located. Air in must equal air out to be a balanced system. 

Closing rooms off can un balance a system. Thermostat location will make a system run short or long or just right. Needs to be in the air flow. 

Anyone test duct leakage?

More info would assist with more focus solutions.
